Abstract (English):
Predmet. Problema lecheniya vtorichnogo kariesa postoyannyh zubov ostaetsya, nesmotrya na dostignutyy v poslednie gody progress vosstanovitel'nyh tehnologiy. Aktivnuyu rol' v patogeneze vtorichnogo kariesa igrayut bakterii, obladayuschie proteoliticheskim deystviem, kotorye razzhizhayut kollagenovye volokna v dentinnoy matrice. Aktual'nost' izucheniya citotoksichnosti serebra, vhodyaschego v sostav stomatologicheskogo protravochnogo gelya, prodiktovana klinicheskimi potrebnostyami vrachey-stomatologov. Issledovanie provedeno s cel'yu opredeleniya koncentracii, kotoraya bezopasna i daet terapevticheskiy effekt. Cel' raboty opredelit' in vitro na fibroblastah desny cheloveka citotoksichnost' kolloidnogo rastvora serebra, yavlyayuschegosya komponentom protravochnogo gelya ETCHMASTER Ag. Metodologiya. V eksperimente ispol'zovali liniyu desnevyh fibroblastov cheloveka, poluchennyh vo vremya hirurgicheskogo udaleniya gipertrofirovannoy chasti desny u pacientov s zatrudneniem prorezyvaniya nizhnih zubov mudrosti. V kachestve issleduemogo materiala ispol'zovali nanokolloidnyy rastvor serebra, soderzhaschiy 250 ppm nAg, imeyuschego razmer chastic v diapazone ot 6 do 12 nm. Ego citotoksichnost' ocenivali po standartam: PN-EN ISO 10993-1, 2010 «Biologicheskaya ocenka medicinskih izdeliy: ocenka i testirovanie v processe upravleniya riskami»; PN-EN ISO10993-5, 2009 «Issledovanie citotoksichnosti in vitro”; PN-EN ISO 10993-12, 2012 «Podgotovka obrazca i sravnivaemyh materialov». Rezul'taty. Liniya fibroblastov, poluchennaya neposredstvenno s desny cheloveka, bol'she, chem kommercheskie linii kletok, podhodit dlya testirovaniya citotoksichnosti stomatologicheskoy produkcii, tak kak naibolee blizka citofiziologicheski v usloviyah in vitro k natural'nym kletkam organizma. Iz grafika, predstavlyayuschego zavisimost' ingibirovaniya redukcii MTT ot koncentracii nAg, vidno, chto 9,4 ppm nAg tormozit redukciyu soli tetrazoliya na 50% (EC50) vo vremya inkubacii kletok s ispytuemymi rastvorami nAg v techenie 24 chasov. Vyvody dannogo issledovaniya rastvora nAg pokazali, chto v techenie 24-chasovogo kontakta s kletkami citotoksi-cheskaya koncentraciya sostavlyaet > 8,3 ppm.
