   <abstract xml:lang="ru">
    <p>Предмет. Многие исследователи подчеркивают, что большинство врожденных пороков имеют мультифакториальную природу и, наряду с рядом других заболеваний (новообразования, бронхиальная астма и т. д.), могут служить индикаторами неблагоприятного состояния окружающей среды. В числе ключевых мишеней негативного воздействия экологических и производственных факторов в организме человека выделяют молекулу ДНК.&#13;
Цель. Провести анализ выраженности нарушений целостности ДНК (фрагментации) у детей с врожденной расщелиной губы и неба из регионов с промышленными нефтехимическими экотоксикантами с применением гель-электрофореза одиночных лейкоцитов.&#13;
Методология. Всего было отобрано 60 детей с ВРГН из регионов с промышленными нефтехимическими экотоксикантами, а также 40 детей без соответствующей патологии из тех же регионов, включенных в контрольную группу. Оценку уровня повреждения ДНК проводили на лимфоцитах периферической крови доноров в возрасте 5–12 лет. Степень повреждения ДНК в одиночных лейкоцитах венозной крови определяли щелочным вариантом метода ДНК-комет путем проведения щелочного гель-электрофореза отдельных клеток.&#13;
Результаты. Анализ полученных данных свидетельствует о высоком уровне генотоксического стресса у детей с врожденной расщелиной губы и неба (ВРГН), существенно отличающемся от аналогичных показателей в контрольной группе. Достоверные различия по ряду ключевых параметров (длина хвоста, процент ДНК в хвосте, хвостовой момент) указывают на более интенсивные процессы фрагментации дезоксирибонуклеиновой кислоты у пациентов с врожденными аномалиями. В совокупности эти результаты позволяют предположить, что у данной когорты имеется повышенная чувствительность к неблагоприятным факторам внешней среды, в частности, к токсическим и мутагенным веществам, присутствующим в местах постоянного проживания этих пациентов.&#13;
Выводы. Полученные данные являются новым аспектом в патогенезе врожденной расщелины губы и неба и могут быть использованы для прогнозирования данного порока у детей, проживающих в регионе с промышленными нефтехимическими экотоксикантами.</p>

   <trans-abstract xml:lang="en">
    <p>Subject. Many researchers emphasize that most congenital defects have a multifactorial nature and, along with a number of other diseases (neoplasms, bronchial asthma, etc.), can serve as indicators of an unfavorable state of the environment. Among the key targets of the negative impact of environmental and industrial factors in the human body, the DNA molecule is distinguished.&#13;
Objective. To analyze the severity of DNA integrity disorders (fragmentation) in children with congenital cleft lip and palate from regions with industrial petrochemical ecotoxicants using gel electrophoresis of single leukocytes.&#13;
Methodology. A total of 60 children with congenital cleft lip and palate from regions with industrial petrochemical ecotoxicants were selected, as well as 40 children without the corresponding pathology from the same regions included in the control group. The level of DNA damage was assessed on peripheral blood lymphocytes of donors aged 5–12 years. The degree of DNA damage in single leukocytes of venous blood was determined by the alkaline version of the DNA comet assay by performing alkaline gel electrophoresis of individual cells.&#13;
Results. The analysis of the obtained data indicates a high level of genotoxic stress in children with congenital cleft lip and palate (CLPP), which differs significantly from similar indicators in the control group. Reliable differences in a number of key parameters (tail length, percentage of DNA in the tail, tail moment) indicate more intense processes of fragmentation of deoxyribonucleic acid in patients with congenital anomalies. Taken together, these results suggest that this cohort has an increased sensitivity to adverse environmental factors, in particular, to toxic and mutagenic substances present in their places of permanent residence.&#13;
Conclusions. The obtained data are a new aspect in the pathogenesis of congenital cleft lip and palate and can be used to predict this defect in children living in a region with industrial petrochemical ecotoxicants.</p>
